Nutrivitas®Vitamin B9 (Folate (6S)-5-Methyltetrahydrofolate Calcium) belongs to the folate family of vitamins (vitamin B9, folic acid) and is a coenzyme form of folic acid. Nutrivitas®Vitamin B9 (Folate (6S)-5-Methyltetrahydrofolate Calcium) is the naturally occurring salt-forming methyl derivative form of folic acid. 5-MTHF, also known as levomethylfolate, is the most biologically active and functional form of folic acid and is more easily absorbed than regular folic acid.

In the mid to late 20th century, scientists discovered that natural folic acid (e.g.pteroylglutamic acid) must be converted to 5-methyltetrahydrofolate (5-MTHF) in vivo through a multi-step metabolism before it can be absorbed and utilised, a process that relies on the catalysis of methylenetetrahydrofolate reductase (MTHFR). Early attempts to produce tetrahydrofolate (THF) and derivatives from folic acid by reduction (e.g.sodium borohydride), methylation and other reactions resulted in low purity and poor stability of the products. Research has focused on optimising reaction conditions (e.g. buffer pH, antioxidant addition) to improve the yield and purity of the intermediate THF. In 2000, the research team solved the technical bottleneck of simple decomposition of activated folic acid by combining L-5-MTHF with calcium salts to form stable crystals (L-5-methylcalcium tetrahydrofolate) and using the lattice energy to resist oxidative degradation. This technology has enabled the first large-scale production of calcium L-5-methyltetrahydrofolate, taking it from the laboratory to industrial application.

Production technology description:
●Nutrivitas®Vitamin B9 (Folate (6S)-5-Methyltetrahydrofolate Calcium) was chemically synthesised by mixing folic acid, a reducing agent, a catalyst and an alkaline solvent in a reduction reaction to produce (R,S)-6-tetrahydrofolate. (R,S)-6-tetrahydrofolate was prepared by mixing (R,S)-6-tetrahydrofolate with p-toluenesulphonic acid and an antioxidant, and chiral cleavage was performed to produce p-toluenesulphonate of 6(S)-tetrahydrofolate. L-5-methyltetrahydrofolate was prepared by mixing 6(S)-tetrahydrofolate p-toluenesulfonate with a methylation reagent and an alkaline solvent for a methylation reaction followed by a ring-opening reaction. L-5-methyltetrahydrofolate was reacted with calcium salt to give L-5-methyltetrahydrofolate calcium.

Nutrivitas®Vitamin B9 (Folate (6S)-5-Methyltetrahydrofolate Calcium) is used in scenarios such as megaloblastic anaemia and increased folate requirements during pregnancy, where direct supplementation with active folate bypasses the metabolic barrier of the MTHFR enzyme; as an adjunct treatment for hyperhomocysteinemia, cardiovascular disease and depression, where it reduces disease risk by modulating the methylation cycle; and for the prevention of neural tube malformations (e.g. spina bifida) and recurrent miscarriages. Common dosage forms include tablets, capsules and lyophilised powder for injection.
